The Investigator ProGest Protein Digestion Station (ProGest) is an automated instrument for the enzymatic digestion of proteins contained in pieces of gel after electrophoretic separation. The ProGest performs enzymatic digestion on up to 96 samples simultaneously. It automates the following steps: ⢠Washing of the gel slices with various buffers ⢠Reduction of disulfide bonds at 60C ⢠Alkylation of cystine residues with iodoacetamide ⢠Removal of excess reagents ⢠Equilibration with digest buffer ⢠Addition of digestion enzyme (optional up to 4 enzymes) ⢠Incubation at 37C for digestion ⢠Elution of peptides from the gel matrix Powers up: Touch screen responds as it should (see photos). We ran the machine test and the robotic arm moved up and down in all the wells (see photos). The syringes also moved up and down. We did not test it further.
